COMPLETES $32 Million SERIES C FINANCING
Funds to Advance Novel SMAC Mimetic Cancer Drug Candidate through Phase 2a
Malvern, PA, August 10, 2010 – TetraLogic Pharmaceuticals, a biopharmaceutical company developing novel small molecule drugs to treat cancer, today announced it has completed a $32 million Series C financing with new investor Clarus Ventures leading the round. Also participating in the financing was new investor Hatteras Venture Partners and existing investors Amgen Ventures, HealthCare Ventures, Latterell Venture Partners, Novitas Capital, Quaker BioVentures and the Vertical Group. In conjunction with the financing, Michael Steinmetz, Ph.D., a Managing Director of Clarus Ventures, and Douglas Reed, M.D., a General Partner of Hatteras Venture Partners, have joined TetraLogic’s Board of Directors.
Proceeds from the financing will be used to advance clinical development of TL32711, TetraLogic’s lead drug candidate for the treatment of cancer. TL32711 is a SMAC mimetic that neutralizes the activity of inhibitor of apoptosis proteins (IAPs) that block tumor cell death. TL32711 is being evaluated in a Phase 1 study as a single agent in patients with solid tumors and lymphoma refractory to standard therapies.

TL32711 is a small molecule peptidomimetic of SMAC (an endogenous regulator of apoptotic cell death) that selectively antagonizes multiple inhibitor of apoptosis proteins (IAPs). Apoptosis, a process of programmed cell death, is the primary way that cancer cells are destroyed by standard therapies and also by the body’s biological responses to cancer. IAPs enable cancer cells to resist being destroyed. SMAC mimetics in preclinical studies have been shown to neutralize IAPs, and thus, overcome resistance and enable cancer cell destruction. TL32711 has demonstrated preclinical anti-tumor activity that supports its clinical development for solid tumor and hematological malignancies as a monotherapy and in combination with other anti-cancer therapies.

TETRALOGIC PHARMACEUTICALS INITIATES PHASE 1 CLINICAL TRIAL OF TL32711 IN PATIENTS WITH REFRACTORY SOLID TUMORS OR LYMPHOMA
Malvern, PA – January 8, 2010 - TetraLogic Pharmaceuticals announced today that the Company has completed dosing of the first cohort in a Phase 1 clinical trial of its selective SMAC mimetic, TL32711. The Phase 1 trial is an open-label, dose-escalation study evaluating the safety and tolerability of TL32711 in adults with solid tumors or lymphoma refractory to standard therapies. The study will also assess the pharmacokinetics, pharmacodynamics and anti-tumor activity of TL32711.

TETRALOGIC PHARMACEUTICALS LICENSES EXCLUSIVE RIGHTS TO NOVEL NECROSIS INHIBITORS FROM HARVARD MEDICAL SCHOOL
Malvern, PA – March 9, 2009 - TetraLogic Pharmaceuticals,
a privately held biopharmaceutical company, announced today
that the Company executed a license agreement under which
it acquired exclusive worldwide rights from Harvard University
to commercialize novel necrostatin compounds and related
drug discovery technology developed at Harvard Medical School. Necrostatins
are small molecule compounds that block a programmed cell
death process called necroptosis that would otherwise kill
cells by necrosis. Necroptosis and the potential effect of
necrostatins were originally discovered by Professor Dr.
Junying Yuan, a pioneer and a recognized leader in the field,
and Dr. Alexei Degterev, at the time a member of Dr. Yuan’s
lab at the Department of Cell Biology, Harvard Medical School.
Further improvement of necrostatin compounds was achieved
in collaboration with Dr. Greg Cuny of the Harvard NeuroDiscovery
Center Laboratory for Drug Discovery in Neurodegeneration.
Necrotic cell death occurs in the course of debilitating
acute and chronic conditions including neurological injuries,
neurodegenerative diseases, cardiovascular injuries, liver
and kidney diseases and injuries, burns, transplant rejections
and ocular diseases and injuries. Necrostatins have shown
potent activity in a variety of preclinical models representing
their possible clinical indications. TetraLogic intends
to commence human clinical studies of necrostatins during
mid-2010.

Dr. Pecora is currently in private practice in Hackensack, NJ. In addition, he has spent the last 18 years in various capacities at Hackensack University Medical Center in Hackensack, NJ, currently serving as Chairman and Executive Administrative Director of The Cancer Center, Senior Scientist at the Institute for Biomedical Research, and Attending Physician of Internal Medicine, Member, Section of Hematology-Oncology. During his tenure at Hackensack University Medical Center, Dr. Pecora has also held the roles of Chairman and Director of The Cancer Center; Director, Hematology and Medical Oncology; Chief, Adult Blood and Marrow Stem Cell Transplant Service; and Medical Director, Stem Cell Collection and Storage Service, among others.
Among his numerous other professional positions, Dr. Pecora is co-Founder and co-managing partner of Northern New Jersey Cancer Associates. Malvern, PA – June 12, 2006 – TetraLogic Pharmaceuticals, a privately held company specialized in discovery and development of oncology therapeutics, announced today that it has completed a $36 million Series B private equity financing. Proceeds from the financing will be invested to advance its Inhibitor of Apoptosis Proteins (IAP) antagonist oncology program through initial clinical trials. The financing was led by San Francisco based Latterell Venture Partners. Also included are new investors, Quaker BioVentures, the Vertical Group and Amgen Ventures, and TetraLogic’s existing investors, HealthCare Ventures, PA Early Stage Partners and Kammerer Associates.
TetraLogic Pharmaceuticals is developing small molecules based on discoveries that underpin the design and synthesis ofSmac Mimetics that drive cancer cells towards destruction by apoptosis, or programmed cell death. “The Series B financing and our investor group provide the resources we need to advanceSmac Mimetics into the clinic and build a successful company,” stated John M. TetraLogic recently closed an $8 million start-up financing. The financing was led by HealthCare Ventures LLC of Princeton, New Jersey and PA Early Stage of Wayne, Pennsylvania. PA Early Stage is one of the biotech venture capital firms that received tobacco settlement investment dollars to be invested Pennsylvania life sciences companies. The Tobacco Settlement Investment Board is managed by the Technology Investment Office of the Pennsylvania Department of Community and Economic Development.
